Mice with p38α deletion in DCs have less susceptibility to chronic colitis-induced colon tumorigenesis. Wild-type (WT) and p38αΔDC mice were injected with 10 mg/kg AOM at day 0 and then given three cycles of 5 d of 2% DSS solution (n = 10–11). (A) Weight changes at the end of each DSS cycle and recovery period were recorded. (BG) Mice were killed at day 80. They were analyzed for colon length (B) and tumor numbers in the whole colons (C), were photographed (D), had miniendoscopy of colon tumors (E), and were analyzed for tumor diameters (F) and tumor load (which indicates the sum of tumor diameter per colon; G). Data represent mean ± SEM. Two-way ANOVA with Bonferroni posttest (A) and two-tailed Student’s t test (B, C, F, and G). Results were replicated (n = 3 experiments). *P < 0.05; **P < 0.01; ***P < 0.001; ns, not significant.
