Ability of GSH/GPx4 in lysates of selenium-replete [Se(+)] L1210 cells to (A) reduce and (B) detoxify different ChOOH species versus a PLOOH species. (A) Reaction mixtures contained the same concentration of hydroperoxide (5α-OOH, 6β-OOH, 7α/7β-OOH, or POPC-OOH), GSH, desferrioxamine, EDTA, and cell lysate in Triton-X100/PBS at 37°C. At the indicated times, lipid were extracted and analyzed by HPLC-EC(Hg). Se(−): lysates of selenium-starved cells; (x): controls lacking GSH or lysate. Plotted data are means ± SEM (n=3).
