Two assays of amyloid fibril formation of the reference sequence VL2-8–J1 and its amyloid fibril-inhibiting mutation L75P: ThT fluorescence assay and electron micrographs. The ThT assay shows fluorescence, indicative of formation of amyloid fibrils (A) and electron micrographs that visualize them (B). Upon exposure of VL2-8–J1 to destabilizing conditions, the protein forms amyloid fibrils. In contrast, the mutated variant, VLJ2-8–J1-L75P, under the same conditions does not show an increase in ThT fluorescence or display amyloid fibrils in electron micrographs.
