Citrate operates as redox-equivalent regenerating agent in product synthesis. The capability of citrate to regenerate NADPH in a targeted oxidoreductase reaction was investigated with Ogataea glucozyma CBS 5766 ketoreductase1 (KRED1-Pglu), Lactobacillus brevis alcohol dehydrogenase (LbADH), and Ralstonia sp. alcohol dehydrogenase (RADH) as lyophilized whole cell (LWC) and crude cell extract (CCE) catalysts. Therefore, citrate was provided to regenerate through endogenous tricarboxylic acid cycle enzyme NADPH, which is consumed by the targeted oxidoreductases in the reduction of acetophenone. Only the “negative control purified enzyme” contained the oxidoreductases in purified form instead of LWC/CCE. The “negative control LWC”: contained all compounds except citrate. The reaction setups with “0.5 mM NADPH” contained NADPH instead of citrate. n = 3.
