Effect of RGC-32 silencing on COL1A1, COL4A1, COL5A1, FN1, ACTA2, and nestin mRNAs in astrocytes. Cultured rat astrocytes were transfected with siRGC-32 or siCTR using Lipofectamine 3000 and then stimulated with TGF-β (10 ng/ml) for 18 h. RGC-32 silencing significantly reduced both RGC-32 mRNA (a) and protein expression (b) after TGF-β treatment, as compared to siCTR expression. RGC-32 silencing significantly reduced the TGF-β-induced mRNA expression of COL1A1, COL4A1, COL5A1, FN1 (c), ACTA2, and nestin (d) when compared with that of siCTR. mRNA expression was normalized to 18S RNA, and the results are shown as fold change over siCTR, which was considered to be 100% (a, c, d).
