Positional assembly of enzymes in a polymersome. (A) A mixture of PS-PIAT and anchor is lyophilized with CalB and then dissolved in THF. This mixture is then injected into an aqueous buffer containing GOx, encapsulating it in the inner compartment and subsequently trapping CalB in the polymeric bilayer. A third enzyme, HRP, is immobilized on the polymersomal perimeter though a covalent linkage to an anchor, creating an outer shell of enzymes.
