Inflammation is present in the diabetic bladder and mediated through NLRP3. The effect of diabetes on the induction of inflammation in the bladder was assessed in the presence and absence of NLRP3 using the Evans blue dye extravasation assay described in the Research Design And Methods section. A: The amount of Evans blue dye in the bladder was increased in diabetic (diab) mice compared with nondiabetic (non-diab) (both NLRP3+/+). n = 5 (nondiabetic) and 12 (diabetic). ***P < 0.0001 by Student two-tailed t test. B: Diabetes did not affect the movement of Evans blue into the bladder in the absence of NLRP3. Bars are mean ± SEM. n = 6 (nondiabetic) and 16 (diabetic). ANOVA followed by Tukey post hoc test was also used to compare all groups. No additional significant differences were found.
