Combination of CD4 neoepitope vaccination and local radiotherapy induces a discrete neoepitope – specific T-cell response only against mutated Smc3. CT26-gp70KO tumor bearing mice (n = 2 per group) were treated with 14 Gy local radiotherapy, a 40 µg of a RNA lipoplex based MHC class II neoepitope vaccine or both as (schedule depicted). 1 × 105 isolated CD8+ splenocytes together with 5 × 104 syngeneic bone-marrow-derived DCs were tested for recognition of the matrix peptide pools, 5 × 104 CT26-gp70KO cells or 2 µg/ml Smc3 peptide by IFNγ ELISpot.
