Examples of HIV cure strategies. (A) The “shock-and-kill” strategy aims to reactivate latent provirus followed by killing of activated cells by viral cytopathic effects on the host immune system. (B) The “block-and-lock” functional cure strategy aims to permanently silence latent reservoirs, for instance by LEDGIN-mediated retargeting of integration to sites that are less susceptible to reactivation after interruption of combination antiretroviral therapy (cART).
