Increasing L451P protein concentrations leads to a proportional increase in transcriptional activity, but the mutant never reaches WT levels. U2OS cells were transiently cotransfected with expression vectors encoding PPARγ WT or PPARγ L451P respectively, and a reporter construct harboring the promoter region of CIDEC. Activation of the luciferase reporter, in the absence or presence of 1 μM rosiglitazone, is expressed as fold induction over that with empty vector (EV).
