Figure 3

Sytox Green assays suggest that belinostat and panobinostat promote baseline NETosis as well as both NOX-dependent and -independent NETosis. Neutrophils were treated with HDACis and/or NETotic agonists and Sytox Green fluorescence intensities were then measured at 4 h by using a fluorescence plate reader. (A) Effects of belinostat and panobinostat on baseline NETosis. (B,C) Neutrophils were activated with PMA (B) or LPS (C) in the presence or absence of belinostat or panobinostat. (D,E) Neutrophils were activated with A23187 (D) or ionomycin (E) in the presence or absence of belinostat or panobinostat. The full data spread is indicated with lines and boxes are marked with the mean (+), median and upper and lower interquartile ranges. * p < 0.05 (One-Way ANOVA with Dunnett post-test, n = 5–7). See Supplementary Figure S5 for additional information.
