For each sorted plate, average threshold rate in events (cells) per second was measured. Flow rate for all samples was 1.0. A) Yield was calculated on the basis of percent loss in volume. B) Upon sequencing, the percentage of sorted cells with TCR sequence reads detected was calculated. Threshold rate was plotted against yield (A) or % of cells with TCR (B) and a linear regression applied to the data. Slopes significantly non-zero (p<0.05). R2 displayed on graphs. Data represent sorts using 70 μm nozzle only.
