miR-148a overexpression or TFR1 knockdown affects HCC proliferation. HepG2 and Huh7 cells were transiently transfected with either a miR-148a mimic, TFR1 siRNAs, TFR1 + miR-148a mimic, Empty + miR-148a mimic or respective negative controls. 24 h post-transfection, cells were seeded (5000 cells/well) into a 96-well culture plate. The optical density (OD) of (A,C,E) HepG2 and (B, D and F) Huh7 cells treated with crystal violet was measured at 570 nm for up to 5 days. Experiments were performed in replicates and repeated at least three times. Data represented as mean ± SEM. Two-way ANOVA was applied to calculate statistical signification of growth curves (statistical difference between NC siRNA and TFR1 siRNA 2 is represented as #). *P < 0.05, **P < 0.01, ****P < 0.0001, ^#P < 0.05, ^##P < 0.01.
