Figure 4.

Inhibition of the phosphatidylinositol 3-kinase/AKT serine/threonine kinase pathway reduces the proportion of SP cells in NCI-H929 and NCI-H929/ABCG2+ cell lines. ABCG2 (A) mRNA and (B) protein expression levels were markedly higher in NCI-H929/ABCG2+ cells compared with control cells (NCI-H929/NC) and uninfected NCI-H929 cells (NCI-H929). (C) Representative flow cytometry plots of SP cells before and after treatment with 20 µM LY294002 or 100 nM rapamycin in NCI-H929 and NCI-H929/ABCG2+ cell lines. (D) Proportion of SP cells was significantly higher in the NCI-H929/ABCG2+ cell line compared with in the NCI-H929 cell line. n=3 tests/group. **P<0.01. ABC, ATP binding cassette; ABCG2, ABC subfamily G member 2; NC, negative control.
