Inhibition of PKC activity enhanced RBSDV accumulation in L. striatellus. (a) Activities of PKC kinase in L. striatellus treated with DMSO, Calphostin C or PMA at 24 hpi. The assay was conducted using the PepTag® non-radioactive PKC assay kit (Promega). Each treatment had 30 L. striatellus. (b) Survival rate of L. striatellus injected with Calphostin C, PMA or 1% DMSO (control) was measured by counting the survival insects at 24 hpi. Each bar represents the mean of 30 L. striatellus per treatment ±s.d. (c) Relative accumulation level of RBSDV RNA in Calphostin C-, PMA- or DMSO-injected L. striatellus at 15 d post-RBSDV acquisition. The accumulation was determined through qRT-PCR using a set of P10 specific primers. The bars represent the mean of three biological repetitions per treatment ± s.d. *p < 0.05; **p < 0.01; ***p < 0.001; n.s., not significant (p > 0.05). All the experiments were repeated three times and similar results were obtained.
