Cytotoxic effect induced by montelukast and carfilzomib in the co-culture system of MM cells and BMSCs. (a) U266 cultured with or without patient-derived BMSCs were exposed to bortezomib (2.5nM) for 24 hours and cell death was assessed using annexin V/PI staining. (b) Montelukast and carfilzomib had minimal effect on patient-derived BMSCs (BMSC 1–3) seeded at densities of 1 × 103 per 96-well plate as assessed by CCK-8 assay after treatment for 24 hours. (c-d) U266 and RPMI 8226 cells cultured with or without BMSCs were exposed to montelukast (25μM) or carfilzomib (10nM) separately or combination for 24 hours and cell death was assessed using annexin V/PI staining.
