Notes: (A) Expression of PTPN9 was significantly depressed by PTPN9 shRNA in SW480 cells. (B) The decreased cell viability induced by H₂O₂ treatment was reversed by PTPN9 shRNA in SW480 cells. (C) H₂O₂-induced inhibition of colony formation was attenuated by PTPN9 knockdown. (D and E) Treatment with H₂O₂ led to the activation of caspase-9 and caspase-3, which was repressed by knockdown of PTPN9. (F) PTPN9 knockdown protected against the release of cytochrome c into the cytoplasm in SW480 cells. An unpaired Student’s t-test (for comparisons of two groups) or one-way ANOVA followed by Tukey’s multiple comparison test was used to evaluate statistical significance (*P<0.05).
