Digestive Enzymes in Solid Tissue Disaggregation

ENZYME PURPOSE
Dispase -Breaks down extracellular matrix
-Detaches cell colonies
-Cleaves attachments between cells and extracellular matrix
Collagenase -Breaks down extracellular matrix
-Breaks peptide bonds present in collagen
Hyaluronidase -Breaks down extracellular matrix
-Cleaves glycosidic bonds in hyaluronan
Papain -Degrades proteins which make up tight junctions between cells
DNase-I -Degrades free-DNA
-Prevents cell aggregation
Accutase -Proteolytic, collagenolytic, and DNase activity
TrypLE -Cleaves cell-cell junctions
-Does not alter antigen expression as trypsin would
