Effect of PRMT5 inhibitor DS-437 on targeted therapy. (A) effect of DS-437 combined with anti-erbB2 antibody 4D5 on tumor size in CT26Her2 tumor model. CT26Her2 tumor cells (0.5 × 106) were injected subcutaneously into Balb/c mice that were treated with control PBS, 4D5 (5 mg/kg, twice per week), DS-437 (10 mg/kg, 5 times per week), or both 4D5 and DS-437. Data represent mean + SEM. One way ANOVA with Tukey HSD test was performed to compare the difference in the tumor size of different treatment groups. (B) function of Tregs in tumor bearing mice. Teffs and Tregs of PBS (CR) or DS-437 (DS) treated mouse lymph nodes were collected by sorting as described above. Teffs were labeled with CTV and stimulated with dynabeads CD3/CD28 beads (0.2 μl/well) with or without Tregs as indicated ratios. After 3 days of culture, proliferation of Teffs were analyzed by flow cytometry. The error bars indicate the SD value. *P < 0.05 with control groups calculated by student t-test. (C) TIL were obtained after treatment as in (A) and the cell population analyzed by flow cytometry (4 mice per group, one tumor from each mouse).
