Effect of MG132 on inflammatory cytokine expression in DN rats. MCP-1 mRNA expression was examined by RT-PCR (A), and MCP-1 (B) protein expression levels were determined using Western blot. In DN rats, the levels of MCP-1 were significantly higher than in NC rats and were reduced after administration of MG132 and deguelin for the indicted time. TGF-β1 mRNA expression was examined by RT-PCR (C), and TGF-β1 (D) protein expression levels were determined using Western blot. In DN rats, the relative levels of TGF-β1 were significantly higher than in NC rats and were reduced after administration of MG132 and deguelin for the indicted time. In DN rats, the concentration of urine MCP-1 was significantly higher than in NC rats and was reduced after administration of MG132 and deguelin for the indicted time (E). NC: normal control group; DN: diabetic nephropathy group; MG132: diabetic nephropathy plus MG132 treatment group; Deguelin: diabetic nephropathy plus deguelin treatment group.
