SC-glucose medium mating assay. Strain 3315α (trp1/trp1 lys2/lys2) in the opaque state was used as the mating type tester for cross print mating assays with WT and mutant MTLa strains. This tester was crossed with mutant strains on SC-glucose agar medium at 24°C for 2 days and then replicated on SC-glucose selection medium (Trp− Lys− Ura−) at 30°C for 3 days to detect prototrophic mating products.
