Glucan contribution to biofilm formation. (A) Strains containing a Gtf deletion with and without a deletion of smu_833 were allowed to form a biofilm for 18 h in TYE medium supplemented with 1% sucrose. Biomass was quantified with crystal violet staining, and the optical density at 562 nm was read. Results are representative of those from at least three independent experiments. NS, P > 0.05; *, P < 0.05, **, P < 0.01; ***, P < 0.001. Statistical analysis was done using Student’s t test. (B) Filtered culture supernatants containing eDNA and Gtf proteins were incubated with sucrose overnight and allowed to form cell-free biofilms, which were visualized with fluorescence microscopy. Cascade blue conjugated to dextran was used to visualize glucan, and SYTO9 green was used for visualization of eDNA.
