Rat motor and somatosensory cortex was fixed in paraformaldehyde, dissociated, and stained. Sorting strategy for separating somatostatin (SOM) and calretinin (CR) expressing interneurons is shown from a representative sort. A) All Events are gated based on DAPI. B) The top 5% of SSC-H events are excluded to avoid debris C) SOM(+) cells are gated. D) CR(+) cells are gated. E) Plot of SOM(+), CR(+), and SOM/CR(−) cells. Note the nearly 100% mutual exclusivity between these SOM(+) and CR(+) populations.
