With the development of bone cancer, PWT and PWL were decreased as compared with control rats. Significant mechanical and thermal hyperalgesia appeared seven days after inoculation of cancer cells. The effects of blocking PI3K–mTOR in the dl-PAG on pain responses to mechanical and thermal stimulation were further determined. Blocking respective mTOR and PI3K by infusion of rapamycin and LY294002 into the PAG attenuated hypersensitive pain responses in bone cancer rats. *P < 0.05 versus control rats and bone cancer rats that received infusion of inhibitors.
