Splenic T cells of ApoE–/– mice and ApoE–/––P. gingivalis mice were isolated and stimulated with anti-CD3 and anti-CD28 antibodies for 72 hours. The supernatants were collected. (A) IL-10 (B) and IL-17 in supernatants of T cells were determined by ELISA. (C) IL-6 in supernatants of splenic cells was assessed by ELISA (n = 6 mice per group).(D) Bone marrow cells were isolated from 5–6-week-old ApoE–/– mice, cells were cultured with 20 ng/mL granulocyte-macrophage colony stimulating factor (GM-CSF) and 1 ng/mL IL-4 for 9 days to obtain DCs. DCs were infected with P. gingivalis at multiplicity of infection (MOI) 0, 25 and 100 for 24 hours.
