5 × 10³ cells per well were seeded in 96-well plate in triplicates for different groups. Cells were treated with 5, 15 and 25 uM of Aβ (1–42) for three different time durations 6 h, 15 h, and 24 h. At the end of respective treatments media was discarded and MTT reagent [3-(4,5-Dimethyl-2-thiazolyl)-2, 5-diphenyl-2H-tetrazolium bromide, M5655, Sigma Aldrich] was added to the wells at a final concentration of 0.5 mg/ml and incubated in a 37°C, CO₂ incubator for 4 h for reduction of MTT to purple Formazan crystals. These crystals were dissolved in 200 μl of DMSO and incubated for 15–30 min with gentle shaking. The absorbance of the solution was read at 570 nm and percent viability was calculated.
