The specific SOD1 inhibition induces apoptosis and hinders the formation of cancer cell colonies. (a) After incubation for 24 h with LD100 (0, 20, and 50 μM), cytometric assays of apoptosis are assessed on RWPE-1 and DU145 cells by Annexin V and propidium iodide staining. (b) After transfection with validated 35 pmol siRNA of SOD1 for 24 h, cytometric assays of apoptosis are assessed on HeLa, DU145, and RWPE-1 cells by Annexin V and propidium iodide staining. (c) Exposure to 50 μM LD100 for 2 weeks hinders the formation of HeLa and DU145 cell colonies, whereas the formation of RWPE-1 cell colonies is not obviously affected. Representative results from three independent experiments are shown (a, b).
