OPN (osteopontin) expression was detected in adipose tissue and ASCs (adipose-derived mesenchymal stem cells). (a) The expression of OPN mRNA in adipose tissue from high-fat diet- (HFD-) induced mice relative to the ones from chow diet- (CD-) induced mice. (b) Expression of OPN was detected by real-time PCR and western blot in ASCs from HFD and CD mice. (c) HFD ASCs and CD ASCs (1 × 106 cells) were cultured in 2 ml complete medium for 48 hours. The supernatant was collected and centrifuged at 1000 × g for 4 minutes, and the OPN concentration was measured by ELISA. The OPN content of the adipose tissue (d) and ASCs in overweight and normal human subjects. (e) OPN expression was analyzed by real-time PCR and western blot. Values are means ± SEM. P < 0.05; ∗∗ P < 0.01; ∗∗∗ P < 0.001. All experiments were repeated at least thrice.
