Salmonella-based combination therapy induces suppressive Tregs. Female 7–8 week old NOD mice were treated with 2.5 μg/mouse of anti-CD3 mAb via i.p. injection for 5 days followed by 2 doses of oral combined vaccines. Splenocytes were harvested from the indicated group at day 30 post-vaccination. (A) Representative FACS plots gated on live CD4 T cells indicate the frequency of CD4+CD25+Foxp3+ T cells in the spleens of mice from each group. (B) Quantification the percentages of CD4+CD25+FoxP3+ Tregs in each treated group. Data is presented as means ± SD are from 2 independent experiments. Statistical analysis using one-way ANOVA shows the significance between combined therapy and vehicle group (*p < 0.05; **p < 0.01). (C) in vitro suppression assay of Treg in culture with CD4+CD25 T responder cells and CD3/CD28 beads.
