Relationship between RNA silencing and CpMV1 infection. (A) Northern blotting of CpMV1-infected fungal strains disrupted for RNA silencing-associated genes. CpMV1 was transferred from EP155 to mutants with disruptions of the dcl (Δdcl1 and Δdcl2), agl (Δagl2), and rdr (Δrdr1) genes, as reported earlier (24, 25, 58). CpMV1-infected fungal strains were then subjected to Northern blotting as described for Fig. 4. CpMV1-infected NB631 and virus-free EP155 were included as references. (B) Effects of CpMV1 infection on dcl2 and agl2 transcript levels. Total RNA was isolated from CpMV1-infected EP155, CHV1-infected EP155, and virus-free EP155. dcl2 and agl2 mRNA levels were compared among the three strains.
