Under normal conditions, POC5 is an essential protein for normal cell cycle progression, and this process is a tightly regulated mechanism. In cycling cells, with wtPOC5, POC5 protein is found interacting with several ciliary proteins that assemble before entering to G1 phase. Weak or no interaction of POC5A429V with ciliary proteins (revealed in this study by immunofluorescence, mass spectroscopy, and CO-IP) results in incorrect assembly and cilium retraction.
