Analysis of translation using polysome profiling. HEK-293 cells were either cotransfected with pCMV-NP868R-(G4S)2-K1ERNAP and pK1E-Luciferase (i.e. C3P3-G1 Luciferase), or pCMVScript-Luciferase plasmids. Cell lysates were fractionated by 15–50% sucrose gradient centrifugation as described in Materials and Methods. The absorbance profile of the gradient is shown (top and bottom of the gradients are indicated). 40S and 60S denote the corresponding ribosomal subunits; 80S, monosome.
