Calvarial osteoblasts from 2 animals per genotype were cultured for 6 d in osteogenic conditions. a Differentiation was analyzed by alkaline phosphatase (ALP) staining (Abs = 405 nm) relative to DNA content (Hoechst 33342; 350 nm, 461 nm) and normalized to wild-type ratios. In identical cultures, gene expression was assessed by qPCR (be). The results represent the mean ± S.E.M. and are representative of duplicate experiments.
