Effects of increased bupivacaine concentration formulations on MSC secretion.
Notes: Secretion levels quantified in cell culture supernatants collected from eMSC treated for 24, 48, and 96 hours with 1 mM of bolus, or 1, 7, or 9 mM construct bupivacaine or untreated free MSC controls were normalized to corresponding basal medium controls (0 mM). (A) The baseline IL-6 levels in the control conditions were 3,839.3±583.44, 5,467.5±663.17, and 11,200±3,014.8 for 24, 48, and 96 hours, respectively. (B) The baseline PGE2 levels in the control conditions were 4,900±402.6, 10,744±1,957.2, and 8,198±1,921 for 24, 48, and 96 hours, respectively. (C) Stimulated IL-6 baseline levels in the control conditions were 12,690.2±2,390.2, 19,435.1±2,943.4, and 40,291.5±7,657.4. Statistically different (*P<0.05, **P<0.002, and ***P<0.0001) from all conditions at given time point (+P<0.05) from bolus and media at given time point, and (++P<0.02) all conditions except 7 mM construct at given time point.
