A higher value of η implies a higher Spinach transcription, which means a lower activity of the Cpf1 enzyme. Cpf1 enzymes behave differently when different PAM site locations are used, especially at concentrations of 20 nM and 50 nM. AsCpf1 prefers an internal PAM site while LbCpf1 prefers the distal PAM site.
