CPP Memory Does Not Require dSub Inputs to NAc or dCA1 Inputs to dSub

(A) Dorsal subiculum (dSub) PYRs were transduced with ArchT-GFP, and yellow light was delivered on ArchT-expressing axons in NAc (ONdSub→NAc) using optic fibers inserted bilaterally above the NAc of dSub-transduced (CamKII-Cre or Vglut2-Cre) mice. Each day, mice performed (n = 26 days from six mice) the 1-day CPP task (like in Figure 2B).

(B and C) ArchT-GFP expression in dSub cell bodies (B) and their axons in NAc (C). Wfs1 staining shows caudal portion of hippocampal CA1 adjacent to dSub.

(D) CPP score (mean ± SEM) during pre-test and test (n = 10 light OFF; 16 ONdSub→NAc days). Light was delivered during CPP ONdSub→NAc test (one 10-min pulse; like in Figure 2K).

(E) dCA1 PYRs were transduced with ArchT-GFP, and yellow light was delivered on their axons in dSub (ONdCA1→dSub) using optic fibers bilaterally inserted above dSub. Mice performed each day (n = 23 days from 6 mice) the 1-day CPP task.

(F) dCA1 ArchT-expressing axons in dSub. The inset confocal image shows dCA1 transduced with ArchT-GFP.

(G) CPP score (mean ± SEM) during pre-test and test (n = 12 light OFF; 11 ONdCA1→Sub days). Light was delivered during CPP test (one 10-min pulse over 15 min; like in Figure 2K).
