Bioinformatic pipeline developed for the identification and classification of molluscan and lophotrochozoan pNP and prohormone sequences. (A) The lophotrochozoan and molluscan databases were downloaded from Sequence read archive (https://www.ncbi.nlm.nih.gov/sra), pre-processed, and assembled locally (not shown). (B) Predicted coding sequence regions from genomic data were downloaded and used when available. Identification of the sine-qua-non prerequisites present in the peptide sequences, clustering, and removal of false positive sequences were performed with several bioinformatics tools (e.g., signal, blast, hmmsearch). (C) Orthology assignments and phylogenetic analysis were performed using all-against-all comparisons with psi-blast and blastp as implemented in the CLANS software. (D) Downstream annotation of the identified peptide sequences were aided through multiple sequence alignments, motif identification, and manual inspection.
