EVTs are representative of the cellular composition of primary syngeneic murine tumors. (A) Schematic description of isolation and 3D culture of EVTs from syngeneic murine subcutaneous tumors. Tumors are harvested 4 weeks after implantation. After isolation and processing of tumors, EVTs are cultured in microwells for one day to yield structures of uniform size. (B) Flow cytometry analysis to compare the cellular composition of EVTs to the primary syngeneic tumors. No significant difference observed in percentage of T-cells, macrophages, CAFs and endothelial cells between the two groups. (C) Flow cytometry analysis to determine the cellular composition of EVTs after culture in Matrigel for 5 days.
