Roscovitine reduces glioma cell viability and potentiates the cytotoxic effects of TMZ on glioma cells in vitro. (A) Effect of RSV on cell viability in U87, U373 human glioma cell lines and C6, rat glioma cell line. The cells were treated with increasing concentrations of RSV for 72 h and cell viability was assessed by MTT assay. (B) Effect of TMZ and RSV co treatment on cell viability of U87 and C6 glioma cell lines. Cells were treated first with indicated concentrations of RSV followed by co-treatment with 100 µM TMZ. After 72 h incubation with the drugs cell viability was measured by MTT assay. (C) Long term colony formation assay for U87 and C6 cells after treatment with either TMZ, RSV or TMZ + RSV. The cells were treated with indicated concentrations of drug(s) for 72 h. After that medium was replaced with fresh DMEM supplemented with 10% FBS and cells were allowed to grow for 14 days with media change every third day. On 14th day, colonies were fixed and visualized by staining with 0.05% crystal violet for 30 min. *p < 0.05, **p < 0.005.
