Aberrant expansion of LAMTOR2-deficient B cells in response to B-cell receptor stimulation. Splenic follicular B cells isolated from LAMTOR2fl/fl or LAMTOR2Cd19/Cd19 mice were stained with cell proliferation dye and stimulated with (A) polyclonal anti-IgM F(ab′)2 fragments; (B) anti-IgM F(ab′)2, and anti-CD40 Ab; (C) CpG or (D) anti-CD40 Ab. B cell proliferative response was analyzed by flow cytometry 2 (CpG) or 3 (all other conditions) days after stimulation. Gates in histograms and adjacent numbers indicate the frequency of divided cells. (E) Frequency of IgG+ class switched B cells was measured within divided cells. FACS plots and adjacent chart are representative of two (CpG) or three independent experiments. Bars represent SD.
