ASPD Accumulate Only in Excitatory Neurons
APPswe-transduced neuronal cultures were treated with 75 nM MG132 for 24 h and stained with the indicated antibodies. The ratio of the ASPD-containing neurons among either MAP2/PV or MAP2/Math2 double-positive neurons was quantified using a Yokogawa CQ1 (see Transparent Methods; inset, mean ± SD, n = 6 for MAP2/Math, 24 for MAP2/PV). ASPD were detected by anti-ASPD rpASD1 or mASD3 antibody. GABAergic interneurons are composed of a highly diverse population of inhibitory neurons, which represent about 20% of cortical/hippocampal neurons (Amaral, 1978). For quantification purposes, we used an antibody against PV, because PV-labeled interneurons represent ∼40% of the total number of GABAergic interneurons in the CA1 and CA3 of hippocampus (Ribak et al., 1990) and the PV antibody clearly stains the neuronal cell body.
