Transport of V-ATPases to the ruffled plasma membrane requires binding between the B2-subunit of V-ATPase and microfilaments. The small molecule enoxacin binds B2 and inhibits this interaction. This may account for some or all of the inhibition of osteoclast bone resorption by enoxacin and bis-enoxacin in vitro. This also may account for reduction of orthodontic tooth movement, periodontal-disease induced bone loss, and bone loss after ovariectomy in pre-clinical models.
