Arbidol impairs LASV GP1 dissociation from GP2. (A) Flag-tagged LASV GP from cell lysates of Lamp1 KO HEK293T/17 cells was immobilized on anti-Flag (M2) magnetic beads, which were then treated with 0 or 40 μM arbidol for 1 h at 4°C. The beads were then subjected to a pulse for 1 min at 37°C at the indicated pH in the presence or absence of arbidol. The extent of GP1 dissociation was then determined by Western blot analysis of GP1 in the supernatant and bead-bound fractions. Data are the averages from four experiments. Error bars represent SEMs. (B) Flag-tagged LASV GP, prepared and immobilized as for panel (A), was pretreated with 0 or 40 μM arbidol. The beads were then exposed to pH 6.5 at 37°C for the indicated time, in the presence or absence of arbidol, and the extent of GP1 dissociation was determined as for panel A.
