A family tree of the rhomboid-like superfamily. This schematic, based on existing sequence analysis and functional data, illustrates the relationship shared by iRhoms with the active proteases and the other inactive pseudoproteases of the rhomboid superfamily. Note this is not an evolutionary model. Inset depicts the seven transmembrane topology of iRhoms, comprising a conserved Rhomboid fold (boxed area), an extended cytoplasmic N-terminal tail and a luminal loop (iRHD).
