Effect of alteplase treatment on platelet activation. Platelet activation was assessed by detecting the surface levels of the α-granule glycoprotein P-selectin and by the activation-dependent binding of PAC-1 to platelet αIIbβ3. Human platelets (5 × 105) were incubated with different concentrations of alteplase (1, 10, 100 or 200 μg/ml) for 60 min at 37°C followed by stimulation with collagen-related peptide (10 μg/ml) for 15 min. Platelet activation was analysed by flow cytometry using phycoerythrin-conjugated mouse anti-human P-selectin antibody (a) or fluorescein isothiocyanate-conjugated mouse anti-human PAC-1 antibody (b).
