Figure 5.
MiR-17-DC reduces T cell proliferation and favored Treg expansion. Pretreated BMDCs were added at different densities (10:1, 20:1, 50:1 and 100:1) to T cells for 3 days and incubated with BrdU (24 hours) to quantify proliferation. (A) LPS-stimulated DCs triggered strong T lymphocyte proliferation, whereas this response was suppressed by miR-17-5p pretreatment. (B) Tregs (CD4+CD25+Foxp3+) were decreased by LPS stimulation but was increased by miR-17-DCs. At high concentration, miR-17-DC-stimulated T cells displayed higher Tregs percentage than imDCs. aCompared to imDCs. bCompared to N+LPS. **P<0.01, ***P<0.001.
Abbreviations: BMDC, bone marrow-derived dendritic cell; N, scramble; imDC, immature dendritic cell; LPS, lipopolysaccharide; Treg, regulatory T cells.
