  1. The interaction between ApoB/Cideb and Sec12/Cideb is not competitive. Flag‐tagged Cideb was co‐expressed with apoB‐48 and an increased dose of HA‐Sec12 in 293T cells. Cells were immunoprecipitated with an anti‐Flag antibody, and the levels of co‐immunoprecipitated proteins were detected by IB following SDS–PAGE.
  2. Cideb‐WT and Cideb‐K128A show similar interaction with ApoB. Flag‐tagged Cideb or Cideb‐K128A was co‐expressed with apoB‐48 in 293T cells. Cells were immuno‐precipitated with an anti‐Flag antibody, and the levels of co‐immunoprecipitated ApoB‐48 were detected by IB following SDS–PAGE.
  3. Levels of serum TAG in WT and Cideb −/− mice injected with AAV containing GFP, Cideb‐WT, or Cideb‐K128A (n = 4 per group) under 16‐h fasting condition.
  4. In vivo VLDL‐TAG secretion. Mice mentioned in (C) were injected with Triton WR‐1339 (500 mg/Kg in PBS), and serum samples at indicated time point were collected for TAG analysis.

  6. Serum ApoB levels. Levels of serum ApoB‐48/100 in WT and Cideb −/− mice expressed GFP, Cideb, or Cideb‐K128A after 16‐h fasting and 4‐h Triton WR‐1339 injection. Albumin stained with Coomassie Blue G‐250 was used as loading control.
