Effects of (+)-rhodoptilometrin on the messenger RNA (mRNA) and protein expression of focal adhesion kinase (FAK), fibronectin, and type I collagen in hGF-1 cells. (A) The expression of FAK, fibronectin, and type I collagen mRNA in hGF-1 cells was determined by qRT-PCR analysis after treatment with various dosages of (+)-rhodoptilometrin for 24 h. The relative FAK, fibronectin, and type I collagen mRNA levels were calculated from the ratio of FAK, fibronectin, and type I collagen over that of an internal control GAPDH gene. (B) FAK, fibronectin, and type I collagen expression in hGF-1 cells were evaluated by western blot analysis after treatment with various dosages of (+)-rhodoptilometrin for 24 h. Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) was detected as an internal control. (C) The FAK, fibronectin, and type I collagen levels were quantified by protein band densitometry analysis using ImageJ software and normalized with the level of GAPDH.
