Gating strategy used to identify myeloid subsets in control (tamoxifen treated SP-CWT/WTFlp+/+ or oil treated SP-CI73T/I73TFlp+/− mice) and SP-CI73T/I73TFlp+/− mice 3 d, 7 d and 2 weeks following intraperitoneal tamoxifen administration (250 mg/kg). BALF and enzymatically digested tissue immune cells were gated on CD45+ following exclusion of doublets. Populations were identified based on a modified protocol from Misharin et al., as previously described (9, 20). Alveolar macrophages were gated based SigF+CD11b expression and confirmed using CD11c, CD64; neutrophils (CD11b+Ly6G+); eosinophils (SigF+CD11b+CD11c ); lymphocytes (CD3+CD11b); and immature macrophages (CD11b+CD64Ly6Chi). Insets represent Giemsa stained cytospins of FACS sorted subsets.
