(A) Phosphorylated serines or threonines can be specifically labeled with fluorescent dyes by beta-elimination and conjugate addition17, then sequenced to determine the amino acid positions of the phosphorylated residues within each molecule, as demonstrated in (B) for CTD repeat peptides phosphorylated at either Serine 2 (top panel) or Serine 5 (bottom panel). Histograms in panels A and B each report observations from 49 image fields.
